"title": "Systematic review supports the idea that no alternative test presently replaces elimination diet test and oral food challenge to diagnose canine adverse food reactions.",
"abstract": "Adverse food reactions in dogs are clinically heterogeneous and overlap with canine atopic dermatitis, and diagnosis still relies on an elimination diet test followed by oral food challenge (EDT+OFC). Numerous allergen-specific tests have been proposed, but their diagnostic value remains controversial. Our objective was to identify, synthesize, and critically evaluate the diagnostic tests used or investigated for canine food allergy, with emphasis on their clinical validity, applicability, and agreement with the EDT+OFC reference standard. In accordance with PRISMA (ie, Preferred Reporting Items for Systematic Reviews and Meta-Analyses) 2020 guidelines, Scopus and PubMed were searched without publication-year restrictions, and records were independently screened in duplicate at the title, abstract, and full-text stages. A meta-analysis was not feasible owing to heterogeneity. Fifty-nine studies (1986 to 2025) from 16 countries were included and grouped into elimination/provocation diets and serological, cutaneous, cellular, and molecular tests. The most consistently validated approach remained EDT+OFC. Serological IgG/IgE (and salivary) tests were poorly correlated with clinical outcome; the standardized patch test showed the best cutaneous performance (high negative predictive value), cellular tests (lymphocyte proliferation) showed the closest concordance with the OFC, and molecular/proteomic methods advanced allergen characterization and the understanding of cross-reactivity. No alternative test reliably replaces EDT+OFC. Although patch testing may seem beneficial, its p",

"abstract": "To adjust the reference interval (RI) for a commercially available canine trypsin-like immunoreactivity (cTLI) assay and the cutoff value for the diagnosis of exocrine pancreatic insufficiency (EPI). Lower limit of the RI was calculated based on the fifth percentile of cTLI concentrations in 102 clinically healthy dogs. Questionnaires were sent to clinicians for cases with serum cTLI concentrations ≤ 10 µg/L. Five board-certified small animal internists blinded to serum cTLI concentration individually reviewed questionnaire responses to clinically classify each dog. Dogs were classified as \"EPI\" or \"not EPI\" based on the consensus when ≥ 4 internists assigned the same classification. The cutoff was determined by maximizing the weighted Youden index (1.34 × sensitivity + 0.67 × specificity - 1). Sensitivity was prioritized, as false negatives were considered more deleterious than false positives. The calculated lower RI limit was ≥ 11.8 µg/L. Forty-seven dogs each were classified as EPI and not EPI. The median (range) serum cTLI concentrations were 1.5 µg/L (0.9 to 7.2) for the EPI dogs and 8.3 µg/L (2.2 to 10.0) for the not EPI dogs. The optimal diagnostic cutoff was 5.5 µg/L, with a sensitivity of 95.7% and specificity of 83.0%. Use of the updated RI (≥ 11.8 µg/L) and diagnostic cutoff value for EPI (5.5 µg/L) should reduce false-negative results when using this commercial assay. There has been an upward shift in serum cTLI measurements with this commercial assay, consistent with clinical observations.",

"abstract": "To compare the pharmacokinetics of intact, split, and crushed levetiracetam (LEV) extended-release (LEV-XR) tablets in dogs. We hypothesized that splitting produces a pharmacokinetic profile similar to intact tablets, whereas crushing results in earlier and higher maximum concentrations. Healthy research dogs were administered 500 mg LEV-XR orally as intact, split, or crushed tablets in a randomized crossover design. Plasma LEV concentrations were measured over 12 hours by high-pressure liquid chromatography. Pharmacokinetic parameters were estimated using noncompartmental analysis and supplementary compartmental analysis and compared using linear mixed-effects models. The study utilized 7 Beagles. Compared with intact tablets, crushed tablets resulted in higher maximum concentration (Cmax) values (61.5 vs 46.0 µg/mL; geometric mean ratio, 1.34; 95% CI, 1.08 to 1.65) and lower 8- to 12-hour exposure (30.0 vs 49.8 µg·h/mL; geometric mean ratio, 0.60; 95% CI, 0.47 to 0.77). Duration above the targeted concentration of 10 µg/mL was longer with intact than crushed tablets (10.9 vs 8.5 hours; mean difference, 2.37 hours; 95% CI, 1.02 to 3.72). Exploratory compartmental analysis estimated an earlier time to maximum concentration (Tmax) for crushed compared with intact tablets. No statistically significant differences were observed between intact and split tablets. Splitting LEV-XR tablets did not significantly alter the evaluated pharmacokinetic parameters, whereas crushing resulted in a higher Cmax, an earlier model-estimated Tmax, and reduced late exposure. "abstract": "Studies describing feline myeloma related diseases (MRD) are limited and reported prognostic indicators are inconsistent. Cats more commonly exhibit visceral plasma cell tumours (PCTs) than marrow-predominant disease, suggesting that diagnostic approaches adapted from the International Myeloma Working Group (IMWG) criteria, incorporating extramedullary involvement and myeloma defining events (MDEs), may better capture clinically relevant disease. We sought to evaluate cats with serum protein electrophoresis (SPE) and immunofixation (IF) confirmed M-proteins using IMWG-adapted criteria, determine how often cases satisfy IMWG-adapted versus classic multiple myeloma (MM) definitions, compare outcomes, characterize clinical and laboratory findings and assess prognostic indicators. A retrospective cohort of 20 cats with SPE/IF-confirmed M-proteins and clinical data was included in the study applying IMWG-adapted and classic MM criteria using available clinical records, laboratory data and cytologic or histologic findings. Survival was evaluated using the log-rank test. 14/20 cats met IMWG-adapted MM criteria, whereas 7 cats met classic criteria. Median survival time did not differ between classic MM (226.5 days) and IMWG-reclassified MM (141 days; p = 0.99). Visceral organ involvement was common; splenic PCTs occurred in 71.4% (10/14) and hepatic PCTs in 87.5% (7/8) of sampled organs. Lytic bone lesions were infrequent (3/14). IgG M-proteins predominated (71.4%). No clinicopathologic variable was significantly associated with survival. IMWG-adapted criteria reclassified MM witho",

"abstract": "Canine atopic dermatitis (cAD) is a chronic allergic inflammatory skin disease that requires multimodal management. Allogeneic mesenchymal stromal cells (MSCs) have emerged as a potential therapeutic option. To evaluate the efficacy and safety of an investigational allogeneic adipose-derived MSC product (PK/CM-1) in dogs with cAD. Dogs with mild-to-moderate cAD were enrolled in a single-blind, randomised study and allocated in a 2:1 ratio either to an experimental group (receiving a single intravenous dose of PK/CM-1) or a control group (receiving three monthly subcutaneous doses of lokivetmab). Clinical outcomes were assessed using the Canine Atopic Dermatitis Extent and Severity Index, 4th iteration (CADESI-04) and pruritus Visual Analog Scale (PVAS) scores on Day (D)28 (primary end-point) and D7, D56 and D84. Safety assessments included haematological and biochemical analyses on D84, along with monitoring for adverse events. Thirty-two dogs were included. By D28, both CADESI-04 and PVAS scores had significantly decreased from baseline in both groups. No significant differences between groups were observed on D28 (p = 0.696 and p = 0.088, respectively). Clinical improvement was sustained up to D84. In the experimental group, CADESI-04 and PVAS scores were lower on D84 than on D28 (p = 0.0074 and p = 0.0015, respectively). Two adverse events (one severe and one mild) occurred during PK/CM-1 administration. No clinically significant haematological or biochemical abnormalities were observed. PK/CM-1 demonstrated comparable efficacy and safety to those of lokivetmab over a 3 ",

"abstract": "Rabies vaccination is conducted along the northern border of Peninsular Malaysia, but paired field serological data are limited. We analysed routine programme data from 154 owned dogs sampled immediately before vaccination and 30-83 days later (median, 43 days). Rabies virus-neutralising antibody levels were measured using the fluorescent antibody virus neutralisation test. Paired levels in IU/mL were compared using the Wilcoxon signed-rank test, and benchmark proportions were compared using the exact McNemar test. The proportion with levels ≥ 0.5 IU/mL increased from 72.7% (112/154; 95% confidence interval [CI], 65.2-79.1%) to 98.1% (151/154; 95% CI, 94.4-99.3%; p < 0.001). Three dogs remained below the benchmark. Median levels increased from 1.98 IU/mL (interquartile range [IQR], 0.38-5.92) to 13.5 IU/mL (IQR, 4.5-13.5); the median paired increase was 5.45 IU/mL (bootstrap 95% CI, 3.31-7.25; p < 0.001). Levels increased in 128 dogs, were unchanged in 19 and decreased in seven. Eighty follow-up values equalled the dataset maximum of 13.5 IU/mL, suggesting possible upper-end censoring. Vaccination history identified 99 previously vaccinated dogs and seven recorded as receiving their first vaccination, limiting comparison of these groups. Nearly all dogs retained at follow-up met the serological benchmark. Convenience sampling, incomplete follow-up and uncertain measurement limits restrict inference to this cohort and preclude estimates of population immunity or clinical effectiveness.",

"abstract": "Against the backdrop of iterative upgrades in pet pathogen detection technology, rapid on-site testing (POCT) has become the core technology for on-site identification of deadly pet diseases. Based on the research and development of new materials, intelligent sensors with high sensitivity, fast response, and high design flexibility have demonstrated strong application value and have become an important development direction for the next-generation technology system in the field of pet pathogen detection. Herein, we report two advanced intelligent material-integrated biosensing platforms: a DNA hydrogel-encapsulated glucose amylase-based assay (RC-HGPGA) and a magnetic nanoparticles (MNPs)-based system where single-stranded DNA (ssDNA) serves as a molecular bridge to conjugate MNPs with invertase (RC-MBI). Both systems operate via a cascade reaction: recombinase polymerase amplification (RPA) of target nucleic acids first activates Cas12a nuclease, which then exerts trans-cleavage activity toward the biosensing elements. Subsequent enzymatic hydrolysis generates glucose, whose concentration is quantifiable using a commercial personal glucose meter (PGM). All experimental procedures were conducted at a constant temperature of 37 °C, eliminating the need for complex thermal cycling equipment. Our findings demonstrate that the RC-HGPGA and RC-MBI platforms achieve ultra-sensitive detection of feline panleukopenia virus (FPV) and canine distemper virus (CDV)-two clinically significant pet viruses-with limits of detection (LODs) as low as 10° copies/μL and 101 copies/μL, respecti",
